Methods

Recombinant protein production

We produce proteins on a laboratory scale. Our scientific and technical expertise comprises the full working cycle from the sequencing to the high-purity functional protein:

  • Molecular biological work to generate expression vectors in accordance with the protein’s properties
  • Expression of the proteins in various cell systems (E. coli, yeast, mammalian cells, insect cells) using the batch method or FED batch fermentation (5l scale)
  • Purification via a range of chromatographic methods (affinity chromatography, hydrophobic interaction chromatography, size exclusion chromatography, ion exchange chromatography, reversed phase chromatography)
  • Associated protein analytics for quality assurance 

Analyses on substrate metabolism and its inhibition

  • Based on spectrophotometric, fluorometric or fluorescence polarisation measurements in the 96 or 384 well format
  • Parameters, such as Km, kcat, Ki, IC50
  • Examinations on the inhibition mechanism of inhibitors
  • Determination of substance solubility using nephelometry

Aggregation experiments

  • Analysis of amyloidogenic proteins using fluorometric methods (Thioflavin T-assay)
  • Statistical analysis of aggregation kinetics (to determine the lag-phase time and aggregation rate)
  • Aggregation visualisation (together with cooperation partners) via transmission electron microscopy (TEM) and atomic force microscope (AFM)
  • Analyses of the inhibition of aggregation by various agents (antibodies, small molecules, plant / bacterial extracts)
